STING protein is phosphorylated by TBK1 subsequent to pathogen infection and the binding of dsDNA to the cytoplasmic sensor cGAS. This allows it to bind to IRF3, which in turn stimulates the production of IFN type 1 and other immune responses. output:Mouse Phospho-STING (Ser365) assay principleThe Mouse Phospho-STING (Ser365) assay quantifies the phosphorylation of mouse STING at Ser365. Unlike Western Blot, the assay is entirely plate-based and does not necessitate gels, electrophoresis, or transfer. Two antibodies are employed in the Mouse Phospho-STING (Ser365) assay: one is labeled with a donor fluorophore, while the other is labeled with an acceptor. The first antibody is chosen for its specific binding to the phosphorylated motif on the protein, while the second is chosen for its capacity to identify the protein regardless of its phosphorylation state. The two labeled antibodies are involved in the formation of an immune complex through protein phosphorylation. This results in the donor fluorophore being in close proximity to the acceptor, which in turn generates a FRET signal. Its intensity is directly proportional to the concentration of phosphorylated protein in the sample and serves as a method for evaluating the protein’s phosphorylation status in a no-wash assay format.
