The specimen reacts with antibodies that have been conjugated to colored particles and precoated on the sample pad during the testing process. The antibody-colored particle conjugate will bind to the drug conjugates in the specimen if there are insufficient drug molecules, resulting in the formation of a colored band at the test region of the membrane. Consequently, a colored band is visible in the test region when the urine is negative for the drug. The immobilized drug conjugate on the test region competes with the drug molecules in the urine for limited antibody binding sites if their concentration exceeds the test’s cut-off concentration.
